目的 采用HPLC指纹图谱、化学模式识别和多指标成分含量测定相结合的方法,对不同居群鸡血藤药材进行综合质量研究,为其质量评价提供参考。方法 收集了5个种源地共15个居群的鸡血藤药材样品,采用HPLC指纹图谱进行分析,结合相似度评价、聚类分析、主成分分析和正交偏最小二乘判别分析进行不同居群鸡血藤药材质量评价,并测定原儿茶酸、儿茶素、表儿茶素、染料木素和芒柄花素5种指标性成分的含量,同时按照《中国药典》的热浸法测定不同居群鸡血藤样品的醇溶性浸出物。结果 15个居群鸡血藤药材共标定了13个共有峰,经对照品比对指认了8个成分,指纹图谱相似度范围为0.692~0.984,具有较好的相似性;聚类分析将样品分为3类;主成分分析用3个主成分进行评价,综合得分显示,产自广东梅州、广西贺州和百色的鸡血藤质量较好;正交偏最小二乘判别分析筛选了不同产地鸡血藤药材的6个质量差异标志物,分别为芒柄花素、毛蕊异黄酮、染料木素、大豆苷元、色谱峰2和色谱峰3。含量测定结果表明不同产地鸡血藤药材中5种指标性成分含量存在明显差异;5种指标性成分的总含量与主成分综合得分存在明显相关性。结论 将HPLC指纹图谱、化学模式识别和多指标成分含量测定相结合的方法不仅可为鸡血藤药材质量控制的综合评价提供参考,也为其种质资源的品质评价提供了一定的科学依据。
Abstract
OBJECTIVE To study comprehensively the quality of Spatholobus suberectus Dunn from different populations based on the methods of HPLC fingerprint, chemical pattern recognition and quantitative analysis of multicomponents, to provide reference for its quality evaluation. METHODS The samples of Spatholobus suberectus Dunn from 15 populations in 5 provenances were collected and analyzed by HPLC fingerprints. The quality of Spatholobus suberectus Dunn from different populations was evaluated by similarity evaluation, cluster analysis, principal component analysis and orthogonal partial least squares discriminant analysis, and the contents of protocatechuic acid, catechin, epigallocatechin, genistein and fermononetin were determined. At the same time, the ethanol-soluble extract of Spatholobus suberectus Dunn samples from different populations were determined according to the hot soak method of Chinese Pharmacopoeia. RESULTS Thirteen common peaks were identified in 15 populations of Spatholobus suberectus Dunn, and 8 components were identified by reference substances. The similarity of fingerprint was 0.692-0.984, indicating good similarity. The samples were divided into 3 groups by cluster analysis; principal component analysis results extracted 3 principal components. According to the composite score, the quality of Spatholobus suberectus Dunn from Guangdong Meizhou, Guangxi Hezhou and Guangxi Baise were better than others. And 6 kinds of marker compounds that cause the quality difference of Spatholobus suberectus Dunn were screened out through the orthogonal partial least square discriminant analysis, which were fermononetin, calycosin, genistein, daidzein and peaks 2 and 3 respectively. The content determination results showed that there were significant differences in the contents of 5 index components in Spatholobus suberectus Dunn from different places, and there was a significant correlation between the total content of the 5 index components and the comprehensive score of the principal components. CONCLUSION The method of combining HPLC fingerprint, chemical pattern recognition and quantitative analysis of multicomponents can not only provide a reference for the comprehensive evaluation of the quality control of Spatholobus suberectus Dunn, but also provide a certain scientific basis for the quality evaluation of its germplasm resources.
